    A comparison of proline, thiol levels and GAPDH activity in cyanobacteria of different origins facing temperature-stress

    Three cyanobacterial strains originating from different habitats were subjected to temperature shift exposures and monitored for levels of proline, thiol and activity of glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Thermophile Mastigocladus laminosus (growth optimum, 40°C), raised the proline level 4.2-fold at low temperature (20deg;C), for the psychrophile Nostoc 593 (growth optimum, 20°C), it was raised 8-fold at 40°C while in the mesophile Nostoc muscorum (growth optimum, 30°C), the imino acid level increased 2.3-fold during temperature 'shiftdown' to 20°C or 3.5-fold in sets facing 'shiftup' (40°C). Alterations in thiol levels in the above strains were in line with proline. It is suggested that such fluctuations reflect metabolic shifts as a response to stress. Interestingly, GAPDH activity was maximum at the respective growth temperature optimum of M. laminosus (122 nmol NADPH oxidized min-1 mg-1 protein) and Nostoc 593 (141 nmol NADPH oxidized min-1 mg-1 protein) while in N. muscorum, it increased at 40°C (101 nmol NADPH oxidized min-1 mg-1 protein) and to 93.3 nmol NADPH oxidized min-1 mg-1 protein (20°C) relative to 86 nmol NADPH oxidized min -1 mg-1 protein at 30°C. It seems that extremophiles maintain the GAPDH activity/level during growth at their respective temperatures optimal while the mesophile increases it in order to cope up with temperature-stress.

    Ab initio evaluation of oxidation numbers in some substituted benzenes

    Giambiagi's definition of oxidation number of an atom in a molecule has been applied successfully in the ab initio SCF theory to calculate oxidation numbers using STO-3G and 4-31G basis sets for some substituted benzenes involving -F, -OH, -CH3 and -NH2 as substituents. The present study suggests that the oxidation numbers also seem to be indicative of their orientational behaviour like the net atomic charges. © 1988 Indian Academy of Sciences.

    Assesment of bioactivity of a Fischerella species colonizing Azadirachta indica (neem) bark

    In order to investigate whether a. Fischerella sp. colonizing Azadirachta indica (neem) bark is able to show, like neem derived extracts/products, bioactivity with respect to glucose metabolizing potential in animal models, laboratory cultivated neem colonized cyanobacterial biomass was fed to mice and its effects on liver lactate dehydrogenase (LDH: a key enzyme of glucose metabolism) vis a vis blood glucose level was monitored. Neem bark extract and purified azadirachtin (a bioactive neem product) were also tested simultaneously for comparison. As compared to the control, none of the samples could produce much variation in blood glucose levels except a transitory decline, 34% and 26% at 2nd & 12th h, in azadirachtin and Fischerella (isolated from neem) fed mice respectively. However, all the test samples showed significant increase in liver LDH activity after 2nd h of treatment (p< 0.01) followed by a decline at 6th h. Thereafter, the enzyme activity increased progressively up to 24th h in mice fed with azadirachtin and neem colonized Fischerella only (p < 0.001 at 24th h). Expression pattern of the enzyme was also studied using polyacrylamide gel electrophoresis. Liver specific M4-LDH (homo-tetramer of M sub-unit) was detected in all the cases and time-dependent changes in expression of the enzyme paralleled the alterations observed in its activities. To ascertain whether such a bioactivity is common with Fischerella sp. colonizing other trees also, liver LDH profile was compared between mice fed with Fischerella isolated from neem and jamun (Eugenia jambolana) barks separately. Though Fischerella from jamun did not alter liver LDH profile, Fischerella isolated from neem could produce significant increase in the enzyme activity (p < 0.001) and corresponding changes in the expression of LDH isozymes. The results suggest that Fischerella sp. inhabiting neem bark, like bioactivity of its host plant products, modulates activity and expression of liver LDH in mice. © 2005, Penkala Bt.

    Assessing a suitable extractant and critical limits of nickel in soil and plant for predicting the response of barley (Hordeum vulgare L.) to nickel grown in Inceptisols

    Nickel (Ni) is an essential element for plants. Abundant information exists on Ni toxicity in soil–plant system but not much is available on its critical level of deficiency (CLD) in soils and plants. Five chemical extractants were evaluated to find a suitable extractant for Ni in Inceptisol. Twenty-one soils having low to high levels of Ni were used to grow barley (Hordeum vulgare L). The amount of Ni extracted was correlated with Ni concentration and uptake by barley. The diethylene triamine penta acetic acid-calcium chloride (0.005 M DTPA-CaC12) was identified as the most promising soil extractant for Ni. The CLD of Ni for 0.005 M DTPA-CaC12 in soil was 0.22 mg kg−1 whereas in barley plant it was 2.14 mg kg−1. Application of 7.5 mg kg−1 Ni in soil caused a significant increase in Ni concentration in the shoot of barley in all the soils irrespective of the initial Ni status. © 2018, © 2018 Taylor & Francis.

    Botulinum toxin in management of neuroma pain: Case report

    Neuroma is a painful condition and always a challenging situation for pain clinicians. Botulinum toxin (BoNT) is a neurotoxin and it provides analgesia, possibly by inhibiting acetylcholine release or blocking autonomic pathways. We report two interesting cases of neuroma in which botulinum toxin (BoNT) serotype-A was locally infiltrated (10 units into the trigger point), three times after an interval of one month each. We observed relief of pain after 10 days and a complete remission after 4 months. © 2006 VSP.

    Cervical epidural anesthesia: A safe alternative to general anesthesia for patients undergoing cancer breast surgery

    Background: General anesthesia (GA) is the standard anesthesia for patients undergoing modified radical mastectomy (MRM) for breast cancer. Cervical epidural anesthesia (CEA) is practiced less often because of its reported complications. This prospective study aimed to evaluate the safety and efficacy of CEA as an anesthetic technique for MRM. Patients and Methods: Fifty breast cancer patients with ASA (American Society of Anesthesiologists) grade I or II underwent MRM under CEA from September 2004 to January 2006. Anesthesia was induced with 10 ml of 1% lignocaine; adrenaline was administered through an 18-gauge catheter in C6-C7 or C7-T1 epidural space. Postoperative analgesia was maintained with 0.125% bupivacaine through the epidural catheter. Results: In 49 (98%) patients surgery was conducted smoothly under CEA with good analgesia. 44 patients were awake during surgery. Five patients had to be given intravenous sedation with midazolam, and in one case the procedure was terminated after accidental dura puncture. There were no clinically significant variations in perioperative pulse and respiratory rate, and there was no fall in mean arterial blood pressure during the procedure. The mean preoperative anesthesia time and total cost of the procedure was 20.36 + 2.75 minutes and 12.19 + 2.2£, respectively. All patients were started on a liquid diet and mobilized 4 hours after surgery. Conclusions: Cervical epidural anesthesia is a safe alternative to GA and was preferred by our patients because of its lower cost and reduced perioperative morbidity. © 2006 Société Internationale de Chirurgie.

    Comparative Evaluation of Oxygen-Ozone Therapy and Combined Use of Oxygen-Ozone Therapy with Percutaneous Intradiscal Radiofrequency Thermocoagulation for the Treatment of Lumbar Disc Herniation

    Aim: To compare the efficacy of oxygen-ozone therapy and the combined use of oxygen-ozone therapy with percutaneous intradiscal radiofrequency thermocoagulation (PIRFT) for the treatment of contained lumbar disc herniation. Methods: Ninety-one adult patients with low back pain secondary to contained lumbar disc herniation were randomly assigned into two groups. Ozone group received intradiscal oxygen-ozone therapy (4 to 7mL of oxygen ozone mixture); ozone-PIRFT group received a combination of oxygen-ozone therapy with PIRFT (radiofrequency lesioning at 80°C for 360s). Outcome Measures: Primary outcome measures included a visual analog scale (VAS) for pain and the Oswestry disability index (ODI). Secondary outcome measures included pain relief, reduction of analgesic consumption, and patient's satisfaction. Clinical assessment of these outcome measures was performed at 2 weeks, 1 month, 3 months, 6 months, and 1 year after the procedure. Results: VAS scores and ODI were significantly decreased by both ozone and ozone-PIRFT when compared with the baseline values at all points of follow-up; however, ozone-PIRFT produced a significant reduction in the VAS scores and ODI when compared to ozone at 2 weeks, 1 month, 3 months, 6 months, and 1 year follow-up. Ozone-PIRFT also resulted in a significant change in all secondary measures at all points of follow-up, as compared with the ozone group. Conclusion: Ozone-PIRFT is more efficacious than ozone alone in reducing pain scores, analgesic consumption, improving functional outcome, and satisfaction of patients with contained lumbar disc herniation. © 2010 World Institute of Pain.

    Comparison of neural networks techniques to predict subsurface parameters based on seismic inversion: a machine learning approach

    Seismic inversion, complemented by machine learning algorithms, significantly improves the accuracy and efficiency of subsurface parameter estimation from seismic data. In this comprehensive study, a comparative analysis of machine learning techniques is conducted to predict subsurface parameters within the inter-well region. The objective involves employing three separate machine learning algorithms namely Probabilistic Neural Network (PNN), multilayer feedforward neural network (MLFNN), and Radial Basis Function Neural Network (RBFNN). The study commences by generating synthetic data, which is then subjected to machine learning techniques for inversion into subsurface parameters. The results unveil exceptionally detailed subsurface information across various methods. Subsequently, these algorithms are applied to real data from the Blackfoot field in Canada to predict porosity, density, and P-wave velocity within the inter-well region. The inverted results exhibit a remarkable alignment with well-log parameters, achieving an average correlation of 0.75, 0.77, and 0.86 for MLFNN, RBFNN, and PNN algorithms, respectively. The inverted volumes portray a consistent pattern of impedance variations spanning 7000–18000 m/s*g/cc, porosity ranging from 5 to 20%, and density within the range of 1.9–2.9 g/cc across the region. Importantly, all these methods yield mutually corroborative results, with PNN displaying a slight edge in estimation precision. Additionally, the interpretation of the inverted findings highlights anomalous zones characterized by low impedance, low density, and high porosity, seamlessly aligning with well-log data and being identified as sand channel. This study underscores the potential for seismic inversion, driven by machine learning techniques, to swiftly and cost-effectively determine critical subsurface parameters like acoustic impedance and porosity. © The Author(s), under exclusive licence to Springer-Verlag GmbH Germany, part of Springer Nature 2024.

    Differential sensitivity to tributyltin of cytochrome-containing and cytochrome-deficient cells of Escherichia coli SASX76

    The effect of tributyltin (TBT) chloride on the growth of cytochrome-deficient and cytochrome-containing cells of Escherichia coli SASX76 was examined. The former cells were found to be at least 20 times more sensitive to TBT. It is proposed that the differential sensitivity of these two cell types to the biocide, TBT, may be due to a different mode of energy generation by cytochrome-deficient and cytochrome-sufficient cells. In addition to the energy state, the pH change caused by the presence and absence of cytochromes which occurred during growth also resulted in a differential sensitivity of these cells. © 1985 Birkhäuser Verlag Basel.

    DNase I nick translation in situ on meiotic chromosomes of the mouse, Mus musculus.

    DNase-I-sensitive sites have been located on the meiotic chromosomes of the mouse, Mus musculus, by the in situ DNase I nick-translation method. We find that: (1) of all the cell types studied, pachytene nuclei are the most sensitive to DNase I; (2) in diplotene the nicks occur preferentially in the vicinity of chiasmata; (3) the sex chromosomes are also sensitive to the enzyme despite their transcriptional quiescence; and (4) in the sex bivalent the nicks are primarily observed in the putative region of recombination. We conclude that, in addition to discriminating between the transcriptionally active and inactive states of chromatin, DNase I identifies recombination-specific chromatin changes in meiotic prophase.

    Does trait predict psychological well- being among students of Professional courses?

    With current drive of Positive Psychology, Psychological Well Being (PWB) has been an imperative concept for research and practices since last two decades. Several studies have been conducted to explore the antecedents of PWB. Present study is also a line of effort in this direction. The study was conducted to examine the role of traits (hope and Big five) in PWB among students of professional courses. The data was collected by using The Trait Hope Scale, Big Five Inventory-10 (BFI-10) and Scales of Psychological Well-Being. The results show that hope was significantly positively associated with PWB. Neuroticism was significantly negatively associated with PWB whereas agreeableness was significantly positively associated with PWB. Extraversion, Openness and Conscientiousness, have not significant association with PWB. © Journal of the Indian Academy of Applied Psychology.

    Effect of Long-Term Use of Organics with Inorganic Fertilizers on Yield and Nutrients Uptake under Rice (Oryza sativa)-Wheat (Triticum aestivum) Cropping System in an Inceptisols of Varanasi

    The long-term experiment has been continuing at research farm of the Institute of Agricultural Sciences, Banaras Hindu University, Varanasi since 1985-1986 on rice-wheat cropping system. Field experiment was conducted during Rabi (winter) season of 2016-17. The experiment was conducted with 12 treatments replicated three times employing randomized block design. Out of which 6 treatments comprised with integration of organics with inorganic fertilizer supply system, 4 treatments included only chemical source of nutrients, one was farmer’s practices and one control. The organic sources of nutrients included were (i) Farm Yard Manure (FYM) (ii) Green manure (GM) Sesbania aculeata (dhaincha) (iii) Crop residues (CR) i.e. wheat straw. Chemical sources of nutrients were urea, diammonium phosphate, muriate of potash as per the treatments. The recommended level of N was partially substituted by 25% or 50% N either through FYM or wheat straw or green manure. The result indicated that highest N, P, K and S content and uptake in grain and straw was recorded in plots receiving 50% NPK+50% through FYM. Treatments which included organic matter along with chemical fertilizers showed higher concentration and uptake of nutrients (N, P, K and S). Concentration of nutrient elements in straw and grain increased due to effect of organic matter in case of N, P and K but remained almost unchanged in case of S. All the organic sources were at par for grain yield of wheat. Continued use of organics with inorganic fertilizers in an integrated manner could sustain yields of wheat and maintained adequate supply of nutrients. © 2024, Indian journals. All rights reserved.

    Effect of organic production system on productive and reproductive performance of cattle

    The present investigation was carried out with the objective to study the productive and reproductive performances of dairy cattle under organic farming systems. Total 24 medium to high yielding cattle has been selected for the study, out of which 12 were reared under organic management system and 12 under conventional management system. For cows maintained under organic management system, the animals were fed ad lib quality green fodder grown organically and limited quantity (1.5 to 2.0 kg/cows/day) of concentrate mixture. Production and reproduction performance of animals of both the groups were compared. The results revealed that the reproductive performance was better in cows maintained under organic production management system than their conventional counterparts. The study on milk quality in terms of composition revealed significantly (P<0.05) higher fat % of milk, while lower (P<0.05) protein % of milk for the cows maintained under organic system as compared to those under conventional system. It can be concluded that the organic management system of dairy cows can be beneficial as depicted by better reproductive efficiency of cows with higher fat % of the milk. © 2020 Agricultural Research Communication Centre.
