PublicationArticle Cloning & sequence identification of Hsp27 gene and expression analysis of the protein on thermal stress in Lucilia cuprina(Blackwell Publishing Ltd, 2016) Manish K. Singh; Pramod K. TiwariHsp27, a highly conserved small molecular weight heat shock protein, is widely known to be developmentally regulated and heat inducible. Its role in thermotolerance is also implicated. This study is a sequel of our earlier studies to understand the molecular organization of heat shock genes/proteins and their role in development and thermal adaptation in a sheep pest, Lucilia cuprina (blowfly), which exhibits unusually high adaptability to a variety of environmental stresses, including heat and chemicals. In this report our aim was to understand the evolutionary relationship of Lucilia hsp27 gene/protein with those of other species and its role in thermal adaptation. We sequence characterized the Lchsp27 gene (coding region) and analyzed its expression in various larval and adult tissues under normal as well as heat shock conditions. The nucleotide sequence analysis of 678 bps long-coding region of Lchsp27 exhibited closest evolutionary proximity with Drosophila (90.09%), which belongs to the same order, Diptera. Heat shock caused significant enhancement in the expression of Lchsp27 gene in all the larval and adult tissues examined, however, in a tissue specific manner. Significantly, in Malpighian tubules, while the heat-induced level of hsp27 transcript (mRNA) appeared increased as compared to control, the protein level remained unaltered and nuclear localized. We infer that Lchsp27 may have significant role in the maintenance of cellular homeostasis, particularly, during summer months, when the fly remains exposed to high heat in its natural habitat. © 2015 Institute of Zoology, Chinese Academy of Sciences.PublicationArticle Colonic carcinoma with multiple small bowel perforations mimicking intestinal obstruction(2006) Satyendra K. Tiwary; Manish K. Singh; Rahul Khanna; Ajay K. KhannaBackground: Carcinoma of the colon may present with perforation proximal to the site of malignancy. Caecum is the commonest site of perforation if the ileocecal valve is patent and the jejunal and ileal perforations are very rare. Case presentation: A 35 year male presented with intestinal obstruction. Emergency laparotomy revealed carcinoma of the transverse colon with multiple pinpoint perforations along antimesenteric border of ileum, which were wrapped with omentum, and no peritoneal contamination was present. Extended right hemicolectomy with jejunocolic anastomosis was done. Patient made uneventful recovery in postoperative period and was treated with adjuvant chemotherapy. Conclusion: Patients with colonic carcinoma and incompetent ileocecal valve may present with intestinal perforation. Increased intraluminal pressure and closed loop obstruction may lead to ischemia and perforation of the small bowel. © 2006 Tiwary et al; licensee BioMed Central Ltd.PublicationArticle Heavy and trace metals in carcinoma of the gallbladder(2013) Somprakas Basu; Manish K. ShuklaBackground: Carcinoma of the gallbladder is the commonest malignancy of the biliary tract in northern India The etiologic relation of specific metals (heavy and trace) and their compounds to neoplasia has been a topic of investigation for some time but not adequately described for carcinoma of the gallbladder. The aim of the present study was to evaluate the relation of heavy and trace metals to this malignancy. Methods: The levels of selenium, zinc, copper, manganese, cadmium, chromium, lead, and nickel were estimated in serum, bile, gallstones, and gallbladder tissue using atomic absorption spectrophotometry. The tests were carried out in 30 patients with gallbladder cancer and 30 sex-matched patients with cholelithiasis. Results: Selenium and zinc levels were significantly reduced (p < 0.001) and copper concentration was found to be significantly higher (p < 0.001) in serum, bile, and gallbladder tissue from patients with carcinoma of the gallbladder. Lead, cadmium, chromium, and nickel levels were elevated in serum and bile in patients with carcinoma of the gallbladder. Conclusions: The present study demonstrated a significant decrease in serum, biliary, and tissue levels of selenium and zinc but increased copper, lead, cadmium, chromium, and nickel levels in patients with carcinoma of the gallbladder, indicating a strong relation between the concentrations of these metals and gallbladder cancer. © 2013 Société Internationale de Chirurgie.PublicationArticle Novel conducting lithium ferrite/chitosan nanocomposite: Synthesis, characterization, magnetic and dielectric properties(Elsevier, 2014) Manish Srivastava; Jay Singh; Rajneesh K. Ojha; Madhu Yashpal; Srivastava SudhanshuA study on Lithium ferrite/chitosan nanocomposite (LFCN), easily moldable into arbitrary shapes, as the conducting polymer and ferromagnetic characteristics is presented. The composite material is produced in the presence of Li0.5Cr0.1Fe2.4O4 and Li 0.5Co0.1Fe2.4O4 nanoparticle by ex-situ polymerizations process. Various characterizations techniques have been used to explore the characteristic of the synthesized products. The frequency dependent dielectric properties and electrical conductivity of all the samples have been measured through complex impedance plot in the frequency range of 1 kHz-6 MHz at room temperature. It was observed that in case of (LFCN), fluctuation in value of (ε′) and (ε″) is ceased over the frequency range of 4 Mz which can be attributed to the steady storage and dissipation of energy in the nanocomposite system. Moreover, it is also observed that electrical conductivity of (LFCN) increases with frequency and its value was found to be (0.032-0.048) (ohm-cm)-1 in frequency range of 1 kHz-6 MHz. Due to its low cost, a simple synthesis process and high flexibility, the proposed LFCN may find applications in various types of electronic components. © 2014 Elsevier B.V.
