The catastrophic events comprising geologic activities and anthropogenic activities leave every year a huge amount of nutrient/metal ions in agricultural land/water bodies, they remain in soil for many years and are the potential cause of phytotoxicity or poisoning of the food chain. Generally metal ions are essential in trace amount for the normal development of animals and plants but become toxic when accumulated at higher concentrations. Among the number of practices, phytoremediation was initially proposed as an environmental cleanup technology for the remediation of metal contaminated land. © 2020 Plant Archives. This involves dynamic and reversible modification of serine/threonine residues of different cellular proteins catalyzed by O-linked N- Acetylglucosaminyltransferase and O-linkedN-Acetylglucosaminidase in an antagonistic manner. We report here that decreasing O-GlcNAcylation enhances the viability of neuronal cells expressing polyglutamineexpanded huntingtin exon 1 protein fragment (mHtt). We further show thatO-GlcNAcylation regulates the basal autophagic process and that suppression of O-GlcNAcylation significantly increases autophagic flux by enhancing the fusion of autophagosome with lysosome. This regulation considerably reduces toxic mHtt aggregates in eye imaginal discs and partially restores rhabdomere morphology and vision in a fly model for Huntington disease. This studyissignificantinunravelingO- GlcNAcylation-dependentregulation of an autophagic process in mediating mHtt toxicity. Reiner; Theo VosBackground: While there is a long history of measuring death and disability from injuries, modern research methods must account for the wide spectrum of disability that can occur in an injury, and must provide estimates with sufficient demographic, geographical and temporal detail to be useful for policy makers. The Global Burden of Disease (GBD) 2017 study used methods to provide highly detailed estimates of global injury burden that meet these criteria. Methods: In this study, we report and discuss the methods used in GBD 2017 for injury morbidity and mortality burden estimation. In summary, these methods included estimating cause-specific mortality for every cause of injury, and then estimating incidence for every cause of injury. Non-fatal disability for each cause is then calculated based on the probabilities of suffering from different types of bodily injury experienced. Results: GBD 2017 produced morbidity and mortality estimates for 38 causes of injury. Estimates were produced in terms of incidence, prevalence, years lived with disability, cause-specific mortality, years of life lost and disability-adjusted life-years for a 28-year period for 22 age groups, 195 countries and both sexes. Conclusions: GBD 2017 demonstrated a complex and sophisticated series of analytical steps using the largest known database of morbidity and mortality data on injuries. GBD 2017 results should be used to help inform injury prevention policy making and resource allocation. Regular updates to injury burden assessments are critical. We report Global Burden of Disease (GBD) 2017 Study estimates on morbidity and mortality for all injuries. methods We reviewed results for injuries from the GBD 2017 study. GBD 2017 measured injury-specific mortality and years of life lost (YLLs) using the Cause of Death Ensemble model. To measure non-fatal injuries, GBD 2017 modelled injury-specific incidence and converted this to prevalence and years lived with disability (YLDs). YLLs and YLDs were summed to calculate disability-adjusted life years (DALYs). Findings In 1990, there were 4 260 493 (4 085 700 to 4 396 138) injury deaths, which increased to 4 484 722 (4 332 010 to 4 585 554) deaths in 2017, while age-standardised mortality decreased from 1079 (1073 to 1086) to 738 (730 to 745) per 100 000. In 1990, there were 354 064 302 (95% uncertainty interval: 338 174 876 to 371 610 802) new cases of injury globally, which increased to 520 710 288 (493 430 247 to 547 988 635) new cases in 2017. During this time, age-standardised incidence decreased non-significantly from 6824 (6534 to 7147) to 6763 (6412 to 7118) per 100 000. Between 1990 and 2017, age-standardised DALYs decreased from 4947 (4655 to 5233) per 100 000 to 3267 (3058 to 3505). Interpretation Injuries are an important cause of health loss globally, though mortality has declined between 1990 and 2017. Future research in injury burden should focus on prevention in high-burden populations, improving data collection and ensuring access to medical care. © Author(s) (or their employer(s)) 2020. Despite consumers' demand of such products, their attitudes towards eco-friendly practices can neither be the same for different consumer groups nor can be treated as they all are equally green. The purpose of this study is to operationalize the green market segmentation based on demographic, psychographic and behavioural characterization in the Indian context of green consumerism. Design/methodology/approach: The deductive approach of questionnaire survey method has been adapted to collect the responses from convenience sampling of Indian consumers using the measured constructs concerning to green consumer psychology. The data were analyzed by applying multivariate techniques of confirmatory factor analysis (CFA), cluster and discriminant analysis. Findings: The results revealed that the three distinctive set of consumer groups are evolved as “keen greens”, “moderate greens” and “reluctant greens” based on the eight cognitive variables used in this study, namely environmental concern (EC), perceived environmental knowledge (PEK), perceived consumer effectiveness (PCE), perception of eco-label, perception of eco-brand and environmental advertisements, green purchase intention (GPI) and green purchasing behaviour (GPB) in the Indian context. Research limitations/implications: The research findings may lack its generalizability in the Indian context, as the survey strategy is confined with the most populated territory of India. To provide its robustness, the future studies need more heterogeneous sample across the country. The research findings could provide the key insights into policymakers and enterprises in the framing of marketing strategies to promote green consumerism in the setting of emerging economies. Originality/value: There is dearth of literature concerning to green consumer segmentation based on the “attitude–intention–behaviour” approach in the Indian context. Therefore, the present study endeavours to manifest a holistic description of green consumer classification based on demographic, psychographic and behavioural characterization. HayA double burden of malnutrition occurs when individuals, household members or communities experience both undernutrition and overweight. Here, we show geospatial estimates of overweight and wasting prevalence among children under 5 years of age in 105 low- and middle-income countries (LMICs) from 2000 to 2017 and aggregate these to policy-relevant administrative units. Wasting decreased overall across LMICs between 2000 and 2017, from 8.4% (62.3 (55.1–70.8) million) to 6.4% (58.3 (47.6–70.7) million), but is predicted to remain above the World Health Organization’s Global Nutrition Target of <5% in over half of LMICs by 2025. Prevalence of overweight increased from 5.2% (30 (22.8–38.5) million) in 2000 to 6.0% (55.5 (44.8–67.9) million) children aged under 5 years in 2017. Areas most affected by double burden of malnutrition were located in Indonesia, Thailand, southeastern China, Botswana, Cameroon and central Nigeria. In this study, D. salina strains of the Indian origin isolated from the Sambhar Lake, Rajasthan, were initially cultured optimally at a 0.5 M NaCl level, and these cultures were exposed to 1–2 M NaCl for up to 2 h for transcriptomic analyses. The cells did not accumulate substantial levels of intracellular Na+. Reduced photosynthetic efficiency was observed at 2 h with an Fv/Fm ratio (~0.2) and thereafter reached a maximum on the 10th day. Concurrently, elevated levels of proline and malondialdehyde (MDA) were also detected at higher salt concentrations, suggesting the initiation of a stress response. The transcriptomic study revealed that amino acid biosynthesis pathways, along with pyruvate and carbohydrate metabolism-related genes, were significantly enriched in 0.5 M versus 2 M NaCl-grown D. salina cells at 2 h. Downregulated genes were mainly related to cytoskeletal and microtubule proteins as owing to the loss of flagellar structures at 2 h. There was a change in the carbon flux because of the upregulation of key genes concerned with glyoxylate and TCA cycles, along with fatty acid metabolism, contributing to the remodeling of membrane lipids, thereby supporting membrane integrity. The upregulated differentially expressed genes (DEGs) were validated through qRT-PCR for the photosynthetic apparatus, antioxidative defense, ion homeostasis, and protein translocation and folding. The metabolomic profile under long-term (10th day) acclimation showed the upregulation of secondary metabolites and sterols. However, AM-fabricated components frequently require finishing operations such as abrasive grinding, which causes a different surface characteristic compared to the conventionally manufactured components. Thus, it is essential to study the effect of process parameters and the heat treatment on surface quality of the AM components because these may behave differently to the conventional manufactured components. In this study, surface characteristics of AM samples of Ti-6Al-4V with a cryogenically cooled finishing operation is compared with that of conventionally processed samples. The samples under investigation were fabricated by two different methods, namely, Direct Metal Laser Sintering (DMLS) and conventional processing. The effect of the two processes on surface characteristics, such as microhardness, surface roughness, X-ray diffraction (XRD), and mechanical properties has been studied. The average surface roughness from cryogenic grinding was reduced by 27.25% and 23.15% for the AM, and 30.08% and 29.13% for conventional samples, as compared to dry and moist conditions, respectively. The finished DMLS and conventional samples showed increase of microhardness by 14.07%, 14.27%, 17.54% and 17.48%, 8.06%, 38.68%, in dry, moist, and cryogenic conditions, respectively. In cryogenic conditions, as the DOC and table feed increased, a greater increase in peak broadening was observed. The peak broadening in XRD of finished DMLS components indicates that a significant level of plastic deformation occurred compared to the finished conventionally manufactured Ti-6Al-4V samples, which suggests that the DMLS samples are more sensitive to the finishing process. Compared to dry and moist grinding, cryogenic grinding was found to have the smallest grains in the layer just below the surface. In this study, we synthesize recent advancements in genome editing methodologies applicable to rice and discuss the ethical considerations and regulatory frameworks surrounding the creation of genetically modified crops. Pi54, a dominant blast resistance gene cloned from rice line Tetep, imparts a broad spectrum resistance against various M. oryzae isolates. Many of its alleles have been explored from wild Oryza species and landraces whose sequences are available in the public domain. Its cognate effector gene AvrPi54 has also been cloned from M. oryzae. Complying with the Flor’s gene-for-gene system, Pi54 protein interacts with AvrPi54 protein following fungal invasion leading to the resistance responses in rice cell that prevents the disease development. In the present study Pi54 alleles from 72 rice lines were used to understand the interaction of Pi54 (R) proteins with AvrPi54 (Avr) protein. The physiochemical properties of these proteins varied due to the nucleotide level polymorphism. The ab initio tertiary structures of these R- and Avr- proteins were generated and subjected to the in silico interaction. In this interaction, the residues in the LRR region of R- proteins were shown to interact with the Avr protein. These R proteins were found to have variable strengths of binding due to the differential spatial arrangements of their amino acid residues. Additionally, molecular dynamic simulations were performed for the protein pairs that showed stronger interaction than Pi54tetep (original Pi54 from Tetep) protein. We found these proteins were forming h-bond during simulation which indicated an effective binding. The root mean square deviation values and potential energy values were stable during simulation which validated the docking results. From the interaction studies and the molecular dynamics simulations, we concluded that the AvrPi54 protein interacts directly with the resistant Pi54 proteins through the LRR region of Pi54 proteins. Some of the Pi54 proteins from the landraces namely Casebatta, Tadukan, Varun dhan, Govind, Acharmita, HPR-2083, Budda, Jatto, MTU-4870, Dobeja-1, CN-1789, Indira sona, Kulanji pille and Motebangarkaddi cultivars show stronger binding with the AvrPi54 protein, thus these alleles can be effectively used for the rice blast resistance breeding program in future. © 2019 Sarkar et al.
