Verma; Ram Krishan NegiHexachlorocyclohexane (HCH), an organochlorine pesticide imposes several harmful impacts on the ecosystem. β- and δ-isomers of HCH are highly toxic, persistent, and recalcitrant to biodegradation, slow and incomplete degradation of β- and δ- isomers have been reported in a few strains. We have isolated a strain designated as Sphingopyxis strain MC4 that can tolerate and degrade high concentrations of α-, β-, γ- and δ-HCH isomers. To date, no other Sphingopyxis strain has been reported to degrade β- and δ-isomers. To understand the underlying genetic makeup contributing to adaptations, the whole genome of strain MC4 was sequenced. Comparative genome analysis showed that strain MC4 harbors the complete pathway (lin genes) required for HCH degradation. Genetic footprints such as presence of lin genes on genomic islands, IS6100 elements in close proximity of lin genes, and synteny in lin flanking regions with other strains reflects the horizontal gene transfer in strain MC4. Positive selection and HGT drive the adaptive evolution of strain MC4 under the pressure of HCH contamination that it experienced in its surrounding niche. In silico analyses showed efficient binding of β- and δ-isomers with enzymes leading to rapid degradation that need further validation by cloning and biochemical experiments. © 2023 Elsevier B.V.PublicationArticle Biogenic synthesis of gold nanoparticles using dual extract of tulsi–Vinca for breast cancer tumor regression in mice(Newlands Press Ltd, 2023) Ravi Pratap; Karishma Niveria; Saurabh Kumar Srivastava; Shilpi Chaudhary; Poonam Sharma; Anita K. Verma; Avanish Singh ParmarAim: This work aims to synthesize the gold nanoparticles (GNPs) using a dual extract of tulsi and Vinca (T+V-Gold) for breast cancer tumor regression. Methods: The GNPs were synthesized and characterized for their microscopic, spectroscopic and crystalline properties. Further, the GNPs were investigated for in vitro and in vivo studies for the treatment of the 4T1-induced triple-negative breast cancer murine model. Results: The GNPs for 4T1 tumor-challenged mice resulted in delayed tumor development and lower tumor burden, with T+V-Gold demonstrating the highest prevention of tumor spread. The antitumor effect of T+V-Gold is highly significant in the glutathione family antioxidants glutathione S-transferase and glutathione in tumor tissue samples. Conclusion: The bioefficacy and anticancer outcomes of T+V-Gold nanoformulation can be used as therapeutic agents and drug-delivery vehicles. © 2023 Newlands Press Ltd. The thematic analysis method was employed to analyze interviews with 22 older adults. Severe loss, serious neglect, and cheating were the perceived transgressions of the older adults from old-age homes whereas day-to-day issues characterized the perceived transgressions of the families’ older adults. Pseudo-forgiveness was observed in the older adults residing in old-age homes because of their feelings of helplessness and their fear of losing respect, while affiliation, perceived harm, the realisation of offence, and perceived consequences of punishment were precursors to forgiveness in family-living older adults. Both groups acknowledged the role of offence, intention, relationships, attributions, and commitment to socio-cultural and moral values in forgiveness. © 2024 Taylor & Francis.
