    Copper (II) complexes of azo phenolate ligands: Synthesis, crystal structure, functional mimetic, and anticancer study

    Three novel copper(II) complexes denoted as [Cu(L1)2] 1, [Cu(L2)2] 2 and [Cu4(L3)4] 3, synthesized from azophenoalate ligands (L1: 3,5-di(tert-butyl)-2-hydroxy azobenzene, L2: 4-methoxy-2-hydroxy azobenzene, L3: 2,4-Bis(phenylazo)recorcinol) were subjected to comprehensive characterization. Structural identification via single crystal X-ray analysis displayed a mononuclear architecture for complex 1 and a tetranuclear arrangement for complex 3. These complexes underwent evaluation for their phenoxazinone synthase activities and peroxidase-mimicking through the oxidation of o-amino phenol (OAP) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) in the presence of H2O2 respectively. Furthermore, their cytotoxic effects were investigated in human breast cancer cell line MCF7 and human cervical cancer cell line HeLa, demonstrating noteworthy selective cytotoxicity towards cancer cells with a GI50 < 10 µg/mL. This underscores the potential of these copper(II) complexes as a scaffold for the development of tailored cytotoxic agents. © 2025 Elsevier B.V.
