    Delta variant SARS-CoV-2 infections in pediatric cases during the second wave in India

    Background: During October 2020, Delta variant was detected for the first time in India and rampantly spread across the globe. It also led to second wave of pandemic in India which affected millions of people. However, there is limited information pertaining to the SARS-CoV-2 strain infecting the children in India. Methods: Here, we assessed the SARS-CoV-2 lineages circulating in the pediatric population of India during the second wave of the pandemic. Clinical and demographic details linked with the nasopharyngeal/oropharyngeal swabs (NPS/OPS) collected from SARS-CoV-2 cases (n = 583) aged 0–18 year and tested positive by real-time RT-PCR were retrieved from March to June 2021. Results: Symptoms were reported among 37.2% of patients and 14.8% reported to be hospitalized. The E gene CT value had significant statistical difference at the point of sample collection when compared to that observed in the sequencing laboratory. Out of these 512 sequences 372 were VOCs, 51 were VOIs. Most common lineages observed were Delta, followed by Kappa, Alpha and B.1.36, seen in 65.82%, 9.96%, 6.83% and 4.68%, respectively in the study population. Conclusion: Overall, it was observed that Delta strain was the leading cause of SARS-CoV-2 infection in Indian children during the second wave of the pandemic. We emphasize on the need of continuous genomic surveillance in SARS-CoV-2 infection even amongst children. © 2022

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field. © 2020 Informa UK Limited, trading as Taylor & Francis Group.

    PolSAR calibration and reconstruction of hybrid polarimetric RISAT-1 data for pseudo quad-pol decomposition: A comparison with quad-pol

    A new approach to reconstruction of pseudo quad-polarized data from hybrid polarimetric data has been presented in this research. The algorithm is based on certain assumptions which were validated upon testing the aptness of the results and their comparison with true optical images of the region under study. This involved direct construction of the 3X3 coherency matrix from the 2X1 scattering matrices obtained from the hybrid polarimetric data. The reasonableness of the assumptions were tested by decomposing the reconstructed pseudo quad-pol data using a coherent decomposition mechanism. The data set used in this project was Level-1 FRS-1 Hybrid Polarimetric data and FRS-2 Quad-pol data of RISAT-1. Reliable scattering retrieval from SAR data involves the calibration of the data. Polarimetric calibration was performed on real and imaginary channels of the single look complex SAR data. The newly developed algorithm was implemented on calibrated data. To extract complete information of different scattering elements of any location, second order derivative of scattering matrix is the most suitable and widely used matrix. Coherency matrix of pseudo quad-pol obtained from hybrid polarimetric data using reconstruction algorithm was decomposed using Yamaguchi four component decomposition for scattering information extraction. The obtained surface, double-bounce and volume scattering were compared with the scattering elements of hybrid-polarimetric decomposition, m-alpha and decomposition of quad-pol data of RISAT-1. The comparison revealed that the results obtained were satisfactory and thus the assumptions made during the reconstruction of pseudo quad-pol data were reasonable for specific purposes. Further comparisons of results using different decompositions technique at pixel level comparison can help better understand the aptness of the algorithm. COPYRIGHT © SPIE.     Superior biomaterials using diamine modified graphene grafted polyurethane

    Surface modification of graphene oxide has been performed using diamine moieties with varying chain length and subsequently chemically grafted with long chain polyurethane for wrapping up of graphene sheet with large polymer chains. Functionalization of graphene and its subsequent grafting have been verified through spectroscopic measurements like NMR, FTIR and UV–visible spectroscopy and the uniform dispersion of graphene sheet in polyurethane matrix is achieved. Nanohybrids exhibit better thermal and mechanical responses along with greater self-assembly as compared to pure polymer. Nanometer dimension molecular sheet to gradual increased size of the order of tens of nanometer, hundreds of nanometer to micron scale assembly has been captured through XRD, small angle neutron scattering, AFM and optical microscopy, respectively. Nature of self-assembly associated with stronger interactions sustain the release of embedded drug (anticancerous dexamethasone) from nanohybrid and larger size of inhomogeinities for longer spacer length further sustain the drug release and thereby able to control the release rate of drug by articulating the chemistry of graphene modifications with suitable spacer length of diamine. Biocompatibility of the nanohybrids is verified with cell line studies using human breast cancer cells MDA-MB-231in terms of cell viability, cell adhesion, fluorescence image, reactive oxygen species and mitochondrial tracker measurements indicating better responses of nanohybrid vis-à-vis pure polyurethane. Thus, the control release of the dexamethasone drug from the nanohybrids along with better biological responses clearly suggests a novel biomaterial for the drug carrier. © 2016 Elsevier Ltd
