Publication A coumarin based fluorescent probe enabling nanomolar detection of Zn2+ and Cu2+(Elsevier B.V., 2022) Kumar, Virendra; Diwan, Uzra; Tyagi, Nidhi; Mishra, Rakesh K.; Singh, Manish Kumar; Upadhyay, K.K.A rationally designed fluorescent probe constructed on a coumarin platform (R-1) distinguished sequentially & distinctly two vital ions viz. Zn2+ and Cu2+ through different mechanistic approaches at their nanomolar concentrations. The native dark state (off-state) of R-1 was illuminated (on-state) upon its interaction with Zn2+ and further reverted back to its natural dark state in the presence of Cu2+ in aqueous medium. The structural details of R-1 and its ensemble with Zn2+ were unambiguously elucidated through their single-crystal XRD studies. The single crystals of R-1-Zn2+ with chloride and acetate counter anions displayed a fascinating 1-D co-ordination polymeric supramolecular structures. The strategic incorporation of pyridine �N in R-1 introduced a yet another binding site and consequently modulated its optical characteristics, while the analogous derivative R-3, inspite of having electron withdrawing �NO2 group, (instead of pyridine �N as in R-1 and R-2) was unable to produce any significant sensing response under similar conditions. The corresponding optical responses with Zn2+/Cu2+ were equated with INHIBIT logic- gate function. The polymorphic nature of milk protein fractions and their association with milk production traits, composition and quality has attracted several efforts in evaluating the allelic distribution of protein locus as a potential dairy trait marker. Genetic variants of ?-Lg have highly significant effects on casein number (B > A) and protein recovery (B > A) and also determine the yield of cheese dry matter (B > A). Molecular techniques of polyacrylamide gel electrophoresis and high-resolution accurate mass-spectroscopy were applied to characterize the ?-Lg protein obtained from the Gangatiri breed milk. Sequence analysis of ?-Lg showed the presence of variant B having UniProt database accession number P02754, coded on the PAEP gene. Our study can provide reference and guidance for the selection of superior milk (having ?-LgB) from this indigenous breed that could potentially give a good yield of ?-Lg for industrial applications. A field study was conducted during the rainy season of 2017 to assess the effect of four nitrogen levels in three rice varieties under the transplanted condition at Agricultural Research Farm, Banaras Hindu University, Varanasi, Uttar Pradesh. The experiment was carried out in a split-plot design (SPD) with three replications. The total number of treatments was twelve involving three rice varieties (V1-BPT-5204, V2-Rajendra Kasturi and V3-HUBR 2-1) in main plots and four nitrogen levels (N1-100, N2-120, N3-140, and N4-160 kg ha-1) in sub-plots. Rice variety BPT-5204 recorded significantly higher growth parameters viz. leaf number hill-1(37.17), SPAD value (chlorophyll content 31.83), the higher number of days taken to 50 percent flowering (103 days), maturity (130.17 days), grain yield (50.73 q ha-1), biological yield (112.90 q ha-1) and harvest index (0.45) as well as NPK uptakes by grain (60.96, 13.01 & 10.95 kg ha-1) and straw (29.72, 7.15 & 101.55 kg ha-1) as compared to other varieties. However, straw yield (62.17 q ha-1) was reported higher under HUBR 2-1 as compared to the rest varieties. Among the nitrogen levels, N4-160 kg N ha-1 was recorded significantly higher above the same parameter as compared to other nitrogen levels. However, their detailed functional characterization in tomatoes needs to be investigated comprehensively. In the present study, tomato hybrids subjected to 60 and 80 days of drought stress conditions showed a significant increase in membrane damage and reduced relative water, chlorophyll and proline content. However, hybrids viz., VRTH-16-3 and VRTH-17-68 showed superior growth under drought stress, as they were marked with low electrolytic leakage, enhanced relative water content, proline content and an enhanced activity of enzymatic antioxidants, along with the upregulation of NAC and other stress-defence pathway genes. Candidate gene(s) exhibiting maximum expression in all the hybrids under drought stress were subjected to detailed in silico characterization to provide significant insight into its structural and functional classification. The homology modelling and superimposition analysis of predicted tomato NAC protein showed that similar amino acid residues were involved in forming the conserved WKAT domain. DNA docking discovered that the SlNAC1 protein becomes activated and exerts a stress-defence response after the possible interaction of conserved DNA elements using Pro72, Asn73, Trp81, Lys82, Ala83, Thr84, Gly85, Thr86 and Asp87 residues. However, the progression is plodding, and still, 80% of energy comes mainly from conventional sources in developing countries. The implementation of procedures depends on the political attitudes, political stability, and quality of institutions. India has a diverse political structure ranging from central government to state government to local governments. In the late �80�s, India witnessed a stiff rise in regional and national political parties, which leads more political competition. This paper tries to explain the possible relationship between political competition and CO2 emission in India. With the application of the time series non-linear ARDL (NARDL) model, this study tries to find the asymmetric relationship between political competition and CO2 emission. In our empirical model, we also include other important elements of environmental quality like innovation and fossil fuel consumption. Empirical results show that political competition is asymmetrically related to CO2 emissions in the long run. Fossil fuel consumption and innovation also have a significant relationship with emissions. Many whey proteins transfer the host defense mechanism to the human body. In this assay, electrophoresis followed by a high-resolution mass spectrometry-based proteomic approach has been applied to identify the whey proteome of Indian Jersey crossbreed bovines. RESULTS: Two search engines, MS Amanda and Sequest HT, have shown more than 29 minor proteins. Chromosomal mapping revealed that chromosomes 5 and 9 are expressing maximum proteins in the whey proteome. The principal component analysis, outlier plots, scree plots, score plots, and loading plots were generated to further assess the results. CONCLUSION: The majorly expressed ones are glycosylation-dependent cell adhesion molecule-1, ubiquitin, desmoglein, annexin, glycoprotein, arginase, histones, peroxiredoxin, vimentin, desmin, catenin, peripherin, and 70 kDa heat shock protein. � 2023 Society of Chemical Industry. � 2023 Society of Chemical Industry.Publication Research Data Curation in Academic Institutions: Challenges & Expectations(Defense Scientific Information and Documentation Centre, 2023) Singh, Manish Kumar; Gireesh Kumar, T.K.In the activity of Research Data Curation, the unpublished datasets generated during research are curated for possible reuse in future research by any researcher. Use of curated data, in several cases, may become helpful in avoiding repetition of efforts involved in generation of datasets. A large number of academic institutions in India are actively involved in research in various knowledge areas. Apart from the doctoral and post-doctoral research in academic institutions, academicians are involved in research projects sponsored by public or private bodies; thereby generating sizeable primary and secondary unpublished datasets worthy of curation in a data repository for possible reuse in some other research. For several reasons, curation efforts for research datasets in academic institutions in the country are negligible when compared to such efforts in research institutions. The present work makes an attempt to identify the cause of negligible research data curation efforts in academic institutions of India by uncovering the associated challenges and discusses the expectations from Research Data Repositories of academic institutions. � 2023, DESIDOC.
